一种植物组织原花色素显微观察的冰冻切片方法

摘 要:

本文介绍了一种适合于植物组织原花色素观察的冰冻切片方法。采用酸性香草醛或对二甲氨基肉桂醛溶液对植物组织中的原花色素类物质进行染色, 于–80 ℃冰冻30 min后, –20 ℃下切片。这种方法能够快速观察到原花色素在细胞中的分布, 并为观察植物组织中易溶于丙酮和醇类的物质提供简便和快速的切片技术。

Abstract:

This research provides a new cryo-sectioning technique for visualizing proanthocyanidins-containing elements in plant tissues. The procedure consisted of steps as follows. First, the proanthocyanidins were stained by vanillin acidic solution or p-dimethylaminocinnamaldehyde acidic solution. Then the samples were frozen at –80 ℃ for 30 min and sectioned at –20 ℃. The distribution of proanthocyanidins in cells could be quickly observed after applying this method. This described sectioning method is suitable for visualizing the substances which are more or less soluble in acetone-water and hydroalcoholic mixtures.
